A cysteine protease from papaya fruit that cleaves proteins to support smooth digestion and gastrointestinal comfort.
What it is
Papain is a cysteine endopeptidase derived from the latex of unripe papaya fruit (Carica papaya). It contains a reactive sulfhydryl group that hydrolyzes peptide bonds, amides, and esters across a wide pH range (4.5 to 8.5) and at temperatures up to 60°C. In dietary supplements, standardized papain powder (expressed in USP units or PU) effectively digests animal and plant proteins in both gastric and duodenal environments.
Also known as: Carica papaya enzyme
What it does
- Breaks complex dietary protein molecules down into short peptides and free amino acids.
- Eases digestive heaviness, bloating, and postprandial discomfort following meat or protein shakes.
- Supports gastrointestinal transit and smooth bowel regularity.
Amplified by probiotics
Papain acts as an upstream enzymatic bio-accelerator by breaking complex proteins down into small oligopeptides and bioavailable amino acids. These cleaved protein fragments provide essential nitrogenous nutrients that support the growth and metabolic colonization of beneficial Bifidobacterium and Lactobacillus strains in the intestinal tract. In turn, probiotic microbes ferment unabsorbed peptide residues into beneficial bioactive metabolites while reinforcing the gut epithelial barrier through tight junction protein upregulation. Probiotics also maintain a balanced, slightly acidic luminal pH, optimizing endogenous protease stability, inhibiting putrefactive bacterial overgrowth, and mitigating gut inflammation.
Typical dose
50–300 mg/day
The evidence
Evidence: moderateClinical research shows that papaya-derived papain preparations significantly improve gastrointestinal symptoms such as bloating, constipation, and heartburn in patients with functional digestive disorders (Muss et al. 2013, Neuro Endocrinol Lett; Roxas 2008, Altern Med Rev).
Safety & who should check first
Generally recognized as safe; may cause hypersensitivity reactions in individuals with latex or papaya allergies. Caution advised when co-administered with oral anticoagulants due to mild antiplatelet activity.
